2-氨基嘧啶 用途与合成方法


化学性质  无色针状晶体。熔点127-128℃(124-125℃)。易溶于水,较难溶于乙醚、苯。易升华。


用途  用于有机合成,作医药中间体、生化试剂。

生产方法  以糠氯酸、硝酸胍为原料,经环合、脱羧、脱氯而得。将加入甲醇钠的甲醇溶液及硝酸胍,在40-45℃下于30min内过滤,得游离胍的甲醇溶液。将此溶液加入反应罐中,在95℃30min内加入糠氯酸-甲醇溶液,加毕,于60±1℃保温反应40min,减压蒸馏,蒸出甲醇至一定的数量后停止蒸馏,将反应物加水溶解,在30-35℃以30%盐酸酸化至pH=1,过滤水洗,在95℃干燥,得粗品环合物。将粗品环合物置于升华器内,在150-300℃分段脱羧升华,得2-氨基-5-氯啶[5428-89-7]。环合、脱羧的收率约67.5%。将异丙醇、水苛性钠、锌泥、(氢氧化锌60%)、2-氨基-5-氯嘧啶及锌粉加入反应罐中,在77-80℃回流8h后,先回收低沸点馏出物,在77-100℃回收异丙醇。冷至77-85℃过滤,以热水洗涤滤饼。滤液及部分洗液合并,加入粒状苛性钠,在50℃以下碱析1h。用甲醛酯于70℃以下提取,分出胶状物,用盐酸调节pH=7.5-8,再分出油状物,然后回收甲醛酯,析出2-氨基嘧啶。在40-50℃干燥得成品,收率为85.7%。另一种方法是用盐酸胍和丙炔醛反应。在装有温度计、滴液漏斗、搅拌器的300ml三口烧瓶中,加入90ml浓盐酸,外部用冰盐浴冷却。在搅拌状况下于10℃以下加27g纯度约80%的盐酸胍,待二氧化碳放完后,再冷却到0℃,滴入11.1g粗丙炔醛(折纯品为8.7g),滴加时放热,反应温度随之上升,但掌握不得超过20℃。反应放热停止后,在室温下放置一夜,然后将反应液移入蒸馏瓶中,于35℃左右减压蒸发浓缩,放冷后,加入60ml30%的氢氧化钠溶液,析出生成物2-氨基嘧啶。将此物在60-70℃用1500ml苯提取,苯溶液放冷后用氯化钙干燥。蒸馏除去苯,得产品10.2-10.7g,产率66.5%-70%。熔点125-127℃。
